肿瘤微环境靶向型新一代纳米药物 Nanorobotsfor Tumor Microenvironment Targeting and Regulation 聂广军 国家纳米科学中心,北京中关村北一条11号 niegj@nanoctr.cn Ithas witnessed that the rapid development on precision design and fabrication ofintelligent next generation nanomedicine-medical nanorobots hold the greatpotential to revolutionize the current landscape of drug development. It isalso clear that tumor microenvironment plays critical roles on either promotionor restriction on primary tumor rapid growth and metastasis. Those achievementshave made targeting and regulation of tumor microenvironment viananobiomaterials a feasible and fruitful strategy, to improve the therapeuticoutcomes for cancer treatment. This presentation will feature our recentdevelopment on using DNA and protein based nanorobots as intelligentnanomedicines to regulate tumor microenvironment to block tumor microvessels orre-store the homeostasis of tumor stroma. Roboticmolecular systems have great potential as intelligent vehicles to enable thedelivery of various potent molecules, which otherwise never could be used astherapeutics due to numerous limitations. Yet, achieving in vivo, precisemolecular-level, and on-demand targeting and delivery has proven extremelychallenging. We developed an autonomous nanorobotic system for targeted cancertherapy, programmed to transport molecular payloads and cause on-site tumorinfarction. Given the robust self-assembly behavior, exceptional designability,potent antitumor activity and minimal in vivo adversity, the nanorobotrepresents a promising strategy for precise drug design for cancertherapeutics. 关键词:肿瘤微环境;功能纳米材料;纳米机器
